چکیده
A library of heptapeptides displayed on the surface of filamentous phage M13 was evaluated as a potential source of affinity ligands for the purification of Rhizomucor miehei lipase. Two independent selection (biopanning) protocols were employed: the enzyme was either physically adsorbed on polystyrene or chemically immobilized on small magnetic beads. From screening with the polystyrene-adsorbed lipase it was found that there was a rapid enrichment of the library with “doublet” clones i.e. the phage species which carried two consecutive sequences of heptapeptides, whilst no such clones were observed from the screening using lipase attached to magnetic beads. The binding of the best clones to the enzyme was unambiguously confirmed by ELISA. However the synthetic heptapeptide of identical sequence to the best “monomeric” clone did not act as a satisfactory affinity ligand after immobilization on Sepharose. This indicated that the interaction with lipase was due to both the heptapeptide and the presence of a part of the phage coat protein. This conclusion was further verified by immobilizing the whole phage on the surface of magnetic beads and using the resulting conjugate as an affinity adsorbent. The scope of application of this methodology and the possibility of preparing phage-based affinity materials are briefly discussed. © 2001 Elsevier Science Inc. All rights reserved.

Lactic acid bacteria display a relatively simple metabolism wherein the sugar is converted mainly to lactic acid. The extensive knowledge of metabolic pathways and the increasing information of the genes involved allows for the rerouting of natural metabolic pathways by genetic and physiological engineering. We discuss several examples of metabolic engineering of Lactococcus lactis for the prod...

Halohydrin dehalogenase from Agrobacterium radiobacter AD1 is a homo-tetrameric protein containing three cysteines per 28 kDa subunit. Under oxidizing conditions the enzyme was found to be susceptible to inactivation which could be prevented by the addition of -mercaptoethanol or glycerol. Gel filtration experiments and SDS-PAGE analysis revealed that inactivation coincided with monomerization ...

This study attempted to introduce the bio-processes in the conventional scouring and bleaching preparation of cotton. The scouring with two types of pectinases, acting under acidic and alkaline conditions respectively, was as efficient as the chemical process in terms of obtained adequate water absorbency of the fabrics. The necessity of surfactants application in scouring was outlined. Bleachi...

During the mashing process of brewing, activity of the amylolytic enzymes decays due to the high temperatures used to gelatinise the starch. Because the different enzymes produce different sugars, high temperatures can be exploited to modify the fermentability of resulting worts. This is especially useful when producing low alcohol beers. The expression a.exp(b.t)-c.exp(d.t) (where t is the tem...

In general the biodegradation of nonchlorinated aliphatic and aromatic hydrocarbons is influenced by their bioavailability. Hydrocarbons are very poorly soluble in water. They are easily adsorbed to clay or humus fractions in the soil, and pass very slowly to the aqueous phase, where they are metabolised by microorganisms. Surfactants that increase their solubility and improve their bioavailabi...
